I've heard that Dr. Troy in Vault City is supposed to ask you for a sample of jet. Well, he sure ain't talking to me. What could have gone wrong?

That happened to me too, and ive played with different genders and CH levels, i also never get asked for a sperm sample :-(

In fact i think if you persuade Phyllis to leave VC first, he wont talk to you at all.

You need to get the medical tour (Doctor 75%) and then leave conversation right away. If you already got the tour, you can't get the quest. But you only miss out on $1000, so it's no big deal in the long run. (You can still get the quest in your quest list later on, though not the reward.)
