I don't think it will be the Enclave, even Fallout 3 made the organization deader than dead. It might be cyborgs that'll be the enemy, a cyborg uprising against the Institute or a little bit of both.

People will think I'm just being a snarky bitter asshole about this shit, but it's all out of love for the Fallout series. I see Fallout as one of the best game series out there. It hurts to see it descend to JJ Abrams' Star Trek level where, yeah it might look nice and cool to see some of the iconic stuff again, but ultimately it's just used to tell the same story over and over again to make a shitgazillion dollars.

Sigh...Back on point, I'm not sure Madison Li will appear in this. She was just too much of a nothingy character. But it's Bethesda so...
